UVLS is not the same size as Mark 41, particularly on 055 which have more 9m strike versions of UVLS. It's a much larger scale weapon than Mark 41 and so can be armed with things like YJ-21. One cannot simply run from a Type 055.So in the ought exercise, yes a single Type 055 operating at ~1000km standoff range from Australian coast and with satellite targeting can very much annihilate the entire Australian navy if engagement started within range.
Then the actual reason you'll need to bring multiple is to cover all sides of Australia so they can't simply run.

No, his translation of "Sooner or later we will come to destroy you." was a bit exaggerated.
To be more precise, the Senior Colonel basically said: "Sooner or later we will seize you." The specific word he employed was 收, which could also be translated as "take," but not quite "destroy."
However, prior to making that statement, the Senior Colonel also warned the DPP that they were on the path towards self destruction, so a reasonable argument could be made that he was intimating the destruction of the DPP. However, that's more implicit, and one of any number of possible interpretations of his broader overall statement.
